Role of Adzuki Bean Extract on Obesity-associated Metabolic Disorders

Abstract

The aim of this work was to evaluate the effects of adzuki bean extract on obesity-related parameters, including lipid metabolism, triglyceride accumulation and inflammatory responses, using both in vivo and in vitro systems. For the in vivo study, rats were divided into 4 groups: group C was fed a control diet, group A was fed the control diet with 1% adzuki bean extract, group CF was fed a high fat diet, and group AF was fed a high fat diet with 1% adzuki bean extract. Data showed that adzuki bean extract improved lipid metabolism in both the normal and high-fat diet groups. Group AF exhibited significantly lower total hepatic lipid accumulation (75.9 ± 10.8 mg/liver) compared to group CF (142 ± 27.4 mg/liver). Correspondingly, group AF had significantly higher fecal lipid excretion (59.8 ± 5.35 mg/wet g) compared with group CF (34.9 ± 5.47 mg/wet g). Adzuki bean extract resulted in a dose-dependant reduction in relative pancreatic lipase activity (IC50 value of 12.5μg/ml), and this suggest that reduction of serum TG and hepatic lipid concentrations with concomitant lipid excretion are likely related to inhibitory effect of adzuki bean extract on pancreatic lipase activity. For the in vitro study, incubation of human adipocytes with adzuki bean extract significantly decreased triglyceride accumulation, glycerol phosphate dehydrogenase activity and inflammatory responses without affecting cell viability. Overall, the beneficial effects of adzuki bean extract demonstrated here make it a potential therapeutic natural agent to modulate obesity-associated inflammation and metabolic disorders.
